GLG1 (Golgi complexLocalized Glycoprotein 1, also CFR1, ESelectin ligand1/ESL1, MG160 and Cysrich FGF receptor) is a 150160kD (reducing, 130kD nonreducing) glycoprotein. It is expressed in both Golgi and/or the cell membrane of multiple cell types, including neutrophils (from rodents, not humans), liver stellate cells, neurons, cardiac myocytes, monocytes and bronchial epithelial cells. In the blood, GLG1/ESL1 collaborates with PSGL1 to mediate leukocyte binding to endothelial cell surfaces. PSGL1 initiates leukocyte tethering while GLG1 promotes slow rolling. GLG1 also serves as an intraGolgi receptor for multiple FGFs, including FGF1, 2, 4, 18 and possibly 3, and as a component of an unusual latent TGFbeta complex. Mature human GLG1 is an 1150 amino acid (aa) type I transmembrane protein. It contains a 1116 aa extracellular/luminal region (aa301145) plus a short 13 aa cytoplasmic segment. The extracellular region possesses a 16 aa polyGln segment followed by 16 Cysrich repeats (aa1161101). There are three potential isoform variants, one of which possess a 24aa extension at the Cterminus, a second that couples the aforementioned Cterminal extension to a deletion of aa147157, and a third that contains a 14aa substitution for aa6851179. It is suggested that the longer Cterminus retains GLG1 in the Golgi, while shorter cytoplasmic segments allow for presentation at the cell membrane. Over aa10481145, human and mouse are identical in aa sequence. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 2ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile ddH2O or PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
