Galanin is a 3kD (predicted), secreted member of the Galanin family of peptides. It is co-expressed with differing neuropeptides in a variety of neuron cell types. Galanin affects multiple metabolic processes by binding to one of three GPCRs. GalR1 blocks insulin secretion, GalR2 initiates neurogenesis and GalR3 influences addictive behavior. The human Galanin proform is 11-12kD in size (predicted) and 104aa in length. Proteolytic processing generates a 30aa mature Galanin peptide aa33-62, plus a phosphorylated (Ser117), C-terminal 59aa GMAP fragment that is apparently involved in the processing of noxious stimuli. Once secreted, Galanin can undergo additional proteolytic degradation. Over aa20-123, the human Galanin proform shares 72% aa identity with mouse Galanin proform. Applications: Suitable for use in Flow Cytometry and Immunohistochemistry. Other applications not tested. Recommended Dilution: Flow Cytometry: 2.5ug labels 10e6 cells, Immunohistochemistry: 8-25ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
