Fibronectin is an extracellular matrix component that exists in different alternately spliced isoforms. Fibronectin mediates cell adhesion in its insoluble state, but not as a soluble molecule. Fibronectins play a role in cell adhesion, migration, differentiation, and specific gene expression. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilutions: Flow Cytometry: 10ul labels up to 1x10e6 cells. For intracellular staining, cells must first be fixed with 4% paraformaldehyde buffer and permeabilized with 0.1% saponin saline buffer., Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. Harvest cells and wash twice in saline buffer., 2. Resuspend up to 1x10e6 cells in 0.5ml of cold 4% paraformldehyde fixation buffer and incubate at RT for 10 mins., 3. Following fixation, wash the cells twice in saline buffer, then once in 0.1% saponin saline buffer., 4. After permeabilization, add 10ul of F4215-13B and incubate the cells for 30 minutes at RT in the dark., 5. Wash the cells twice with 0.1% saponin saline buffer., 6. Resuspend the cells in saline buffer for final flow cytometric analysis. As a control for this analysis, cells in a separate tube should be treated with APC-labeled mouse IgG1 antibody. Storage and Stability: May be stored at 4°C before opening. DO NOT FREEZE! Stable at 4°C as an undiluted liquid. Stable for 12 months at 4°C. Freezing allophycocyanin conjugates will result in a substantial loss of activity. Allophycocyanin conjugates are sensitive to light.
