FADD was originally isolated as a protein that bound to the cytoplasmic domain of Fas in the yeast two-hybrid system. Sequence analysis revealed a region homologous to the death domain of Fas and TNFR-1. Subsequent biochemical studies have shown that FADD associates with Fas through interaction of the death domains. When over expressed in several cell lines, FADD induces apoptosis, which can be blocked by CrmA, an inhibitor of Caspase 1 (ICE). This evidence suggests that FADD plays a role in Fas-mediated apoptosis. Applications: Suitable for use in Western Blot. Other applications have not been tested. Recommended Dilutions: Western Blot: 0.5-2ug/ml, detects FADD in RIPA lysated from Jurkat cells, Optimal dilutions to be determined by researcher. Positive Control (included): J9000: Jurkat Cell, Hybridoma: Fusion of mouse myeloma P3U1 with Balb/c splenocytes. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
