E3330-01D is a very useful tool for clinical and experimental allergy studies. BMK-13 is a pan-eosinophil marker in both frozen sections and paraffin-embedded tissues. It stains in bronchial sections, 98% of the peripheral blood eosinophils. Further, the antibody can be used in specific ELISA tests to determine the release of MBP and to quantify the number of eosinophils in various in vitro adhesion assays. This antibody is suitable for blood smears (cytospins) if fixed with methanol-acetone mixture (1:1, v/v). It is unlikely, without permeabilization, that the BMK-13 is suitable for the flow cytometry because BMK-13 stains intracellular, stored proteins. Applications: Suitable for use in ELISA, Immunocytochemistry and Immunohistochemistry. Other applications not tested. Recommended Dilution: Immunohistochemistry (Frozen): 1:30 diluted in PBS using the APAAP method. Incubate sections for 1 hour at RT. , Immunohistochemistry (Paraffin): 1:30 diluted in PBS. Incubate sections for 1 hour at RT. Enzymatic pretreatment for paraffin sections is necessary. Pepsin is recommended. Do not use heat denaturation., Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
