DSC1 (Desmocollin [Greek for "gluethatbinds"]1) is an ~95-110kD member of the Ca++dependent cadherin family of adhesion molecules. It is found on the surface of stratified epithelial cells, including the spinous and granular layers of keratinized and nonkeratinized epithelia of the oral cavity and skin. DSC1 expression is induced by DSG-1. It serves as a component of desmosomes, forming a linkage that unites adjacent cells with cytoplasmic intermediate filaments. In particular, homodimeric DSC1 may form heterotypic interactions with DSG1 intrans, and bind to the cytoskeleton intracellularly via plakophilin1. Mature mouse DSC-1 is a 760 amino acid (aa) type I transmembrane glycoprotein (aa135-691). The mature molecule contains a 557aa extracellular region with five cadherin domains (aa135-682), and a 172aa cytoplasmic domain. There is one splice variant that shows an 11 aa substitution for aa822-886. Over aa135691, mouse DSC-1 shares 82% aa sequence identity with human DSC1. Applications: Suitable for use in Flow Cytometry, ELISA, Western Blot, Immunohistochemistry and mmunocytochemistry. Other applications not tested. Recommended Dilution: Flow Cytometry: 2.5ug/10e6 cells, Western Blot: 0.1ug/ml, Immunocytochemistry: 8-25ug/ml Immersion fixed A549 human lung carcinoma cell line, Immunohistochemistry (Frozen): 1-25ug/ml Perfusion fixed mouse skin, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
