Anti-DISC1 (Disrupted in Schizophrenia 1, RP4-730B13.1, KIAA0457)

Disrupted in schizophrenia 1 (DISC1) is a 100kD cytoplasmic scaffold protein that is associated with the development of schizophrenia, bipolar disorder, and recurrent major depression. It plays a role in post-synaptic density development and neurogenesis as well as centrosome and microtubule dynamics. DISC1 interacts with a range of intracellular proteins including Kinesin 1, NDE1, PDE4, GSK-3 beta, GRB2, PACAP, RAC1, TNIK, and FEZ1. It contains four coiled-coil domains aa366-394, aa452-505, aa602-666, and aa802-830 which mediate the assembly of DISC1 into a variety of multimers. DISC1 multimerization and its ability to interact with various binding parters are regulated by post-translational modifications and proteolysis. Alternate splicing generates additional isoforms of human DISC1 that are truncated before the first or following the third coiled coil domain. Within aa101-260, human DISC1 shares ~43% aa sequence identity with mouse and rat DISC1. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 2ug/ml, Immunohistochemistry: 8-25ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
