Dopamine- and cAMP-Regulated Phosphoprotein, Mr 32kD (DARPP-32), also known as PPP1R1B, is a 23kD protein that anomalously migrates at about 32-35kD on SDS-PAGE. When phosphorylated at T34 by protein kinase A (PKA), DARPP-32 is a potent inhibitor of protein phosphatase 1 (PP1). Dephosphorylation of DARPP-32 at T34 is achieved primarily by the calcium-dependent activation of the phosphatase calcineurin. DARPP-32 is expressed almost exclusively in neuronal tissues, with highest levels in dopamine-innervated neurons. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilutions: Western Blot: 1ug/ml , Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS, 0.02% sodium azide. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
