CD43, a member of the sialomucin family of cell surface receptors, is a 95-135kD type I transmembrane glycoprotein whose variability in molecular weight is dependent upon the nature of the extracellular O-glycans. It is the major sialoglycoprotein on thymocytes, T cells and neutrophils. It is also present on activated B cells, plasma cells, NK cells, granulocytes, monocytes, macrophages, platelets and bone marrow hematopoietic stem cells. CD43, which binds to ICAM-1 (CD54), appears to function as an anti-adhesion molecule, inhibiting T cell interactions, including T cell killing, and by increasing the threshold for T cell activation. Applications: Flow cytometry, Immunohistochemistry (frozen sections and paraffin sections), Recommended Dilution: Flow Cytometry: < 1ug/10e6 cells, Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Store at -20°C or colder. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
