Programmed Death-1 (PD-1) is type I transmembrane protein that belongs to the CD28/CTLA-4 family of immunoreceptors (1). In addition to PD-1, this family also includes CD28, CTLA-4, and ICOS (2-4). Signaling mediated by the CD28/CTLA-4 family regulates immune responses. PD-1 is most closely related to CTLA-4 and shares ~24% amino acid (aa) sequence identity. PD-1 is expressed on activated T cells, B cells, myeloid cells, and on a subset of thymocytes. Binding of PD-1 to PD-L1 or PD-L2 results in the inhibition of TCR-mediated proliferation and cytokine production as well as BCR-mediated signaling (1-4). Mice deficient in PD-1 have a defect in peripheral tolerance and spontaneously develop autoimmune diseases. These observations collectively suggest that PD-1 likely has an inhibitory role in regulating immune responses. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Flow Cytometry: 10ul, 1x10e6 cells, Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C before opening. DO NOT FREEZE! Stable at 4°C as an undiluted liquid. Dilute only prior to immediate use. Stable for 12 months at 4°C. Freezing R-Phycoerythrin (PE) conjugates will result in a substantial loss of activity. PE conjugates are sensitive to light.
