CD152, also known as CTLA-4, is homologous to the costimulatory molecule CD28.1,2 Like CD28, the CD152 molecule binds the family of B7 counter-receptors (CD80/B7-1 and CD86/B7-2) on antigen-presenting cells. CTLA-4 is not expressed on resting T lymphocytes but is induced 2-3 days following T-cell stimulation via the T cell receptor (TCR) complex.1,3,4 Whereas CD28 provides a positive costimulatory signal, CD152/CTLA-4 is a negative regulator of T-cell activation, possibly by inhibiting tyrosine kinase signalling through the TCR through its association with tyrosine phosphatases such as SHP-2.1,5,6, Applications: Flow cytometry 1, Immunoprecipitation1, In vitro and in vivo blocking of CTLA-4 action by soluble antibody 1, Promotion of in vitro CTLA-4 signalling by cross-linked antibody 1, Recommended Dilution: Flow Cytometry: < 3ug/10e6 cells, Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Store at -20°C or colder. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
