Caspase-4, a member of the Caspase-1 subfamily of cysteine proteases, exists as an inactive pro-enzyme which undergoes proteolytic cleavage of its p30 precursor form, into smaller p20 and p10 subunits. The cellular localisation of Caspase-4 on the Endoplasmic Reticulum (ER) membrane, has resulted in this protein becoming a focus of studies in which dysfunction or stress to the ER membrane is implicated, such as Alzheimer's disease and Ischemia (1, 3) and confirms the involvement of Caspase-4 as an instigator of cellular apoptosis. , Applications: Suitable for use in Immunohistochemistry and Western Blot. Other applications not tested. Recommended Dilution: Immunohistochemistry (paraffin): 2ug/ml. Does not require protein digestion pre-treatment or antigen retrieval using heat treatment prior to staining paraffin sections., Western Blot: 1ug/ml. Detects a band at ~34kD in Ramos cell lysates., Optimal dilutions to be determined by the researcher. Positive Control: Spleen tissue, Recommended Secondary Antibody: I1903-30U: IgG, H&L, X-adsorbed (HRP), Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
