This anti-BTLA (B- and T-lymphocyte attenuator) antibody is a purified, recombinant human IgG4 monoclonal antibody that recognizes human BTLA, also known as CD272. This antibody is similar to Venanrpubart. Venanprubart is being studied for the treatment of SLE (Systemic Lupus Erythematosus) and Psoriasis. HVEM (herpesvirus entry mediator), also known as CD270 and TNFRSF14, is a human cell surface receptor in the TNF (tumor necrosis factor)-receptor superfamily that can act as both a co-stimulatory receptor and a co-inhibitory receptor of T cells. Binding of HVEM to one of its ligands, LIGHT (CD258, TNFSF14) or LTalpha (lymphotoxin-alpha), causes a co-stimulatory signal which can activate lymphoid cells. Alternately, interaction with BTLA (B- and T-lymphocyte attenuator), also know as CD272 or CD160, causes a co-inhibitory signal which negatively regulates T-cell immune responses. HVEM has also been shown to interact with adaptor proteins TRAF2 (TNF receptor associated factor 2) and TRAF5, and it is critical to herpes simplex virus (HSV) cellular entry. BTLA and HVEM are considered as immune checkpoints and regulate the TME (tumor microenvironment). Inhibition of this interaction holds great promise in cancer therapy. Antibodies against BTLA, such as Venanprubart, are being studied for the treatment of SLE (Systemic Lupus Erythematosus) and Psoriasis.
