Beclin1 is a 60kD member of the Beclin family of proteins. It is part of a lipid-kinase complex that includes UVRAG and PI3 and induces autophagy, in the presence of Bcl-2, autophagy is repressed. Beclin1 is widely expressed but abundantly found in mammary epithelium, glia and neurons. Human Beclin1 is 450aa in length and contains a Bcl-2 binding domain aa88-150, a nuclear export signal motif aa180-189, a coiled-coil region aa142-270, and an evolutionary conserved domain aa244-337. There is one potential alternate start site at Met88. Over aa12-239, human Beclin1 is 98aa identical to mouse Beclin1. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 0.1ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
