MCA32 (Mast Cell Antigen, also Allergin1 and c17orf60) is a 70kD member of the Ig Superfamily. It is expressed on mast cells, basophils, neutrophils and macrophages, and is reported to negatively regulate allergic mediator release from mast cells. In particular, MCA32 interacts with Fc epsilon RI. Upon IgE mediated Fc epsilon RI aggregation and activation, Allergin1 recruits Tyr phosphatases to a Fc epsilon RI tetrameric complex, dampening IgE receptor downstream signaling. Mature human Allergin1 is a monomeric type I transmembrane glycoprotein that is 324 amino acids (aa) in length. It contains two C2type Ig domains (aa35-118 and 123-217) in the extracellular region (aa20227), plus a 95 aa cytoplasmic domain that possesses two ITIM motifs. Two 40kD splice variants exist that show deletions of the Iglike domains. One isoform termed Allergin1S1 is missing aa128217, while a second isoform called Allergin1S2 is missing aa33122. This second isoform represents the antigen used for immunization in this product (GenBank BAJ08253). Over aa29131, the Allergin1S2 isoform shares 100% sequence identity with human Allergin1L full length isoform and only 24% aa sequence identity with mouse Allergin1. Applications: Suitable for use in Flow Cytometry, ELISA and Immunocytochemistry. Other applications not tested. Recommended Dilution: Flow Cytometry: 2.5ug/ 10e6 cells, Immunocytochemistry: 8-25ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
