Tissue acidosis that occurs in ischemia, tissue damage or inflammation is accompanied by pain. Proton-gated cation channels are activated by low pH in nociceptive neurons. H+-gated channels are members of the NaC/DEG superfamily that include: (1) Amiloride-sensitive epithelial Na+ channels (a, b, and g, and d-ENaC subunits), (2) A FMRFamide-gated channel (FaNaC), (3) and mechanosensory channel proteins of nematode degenerins (DEG). NaC/DEG superfamily is characterized by intracellular N and C-termini, two transmembrane domains, and a large extracellular loop. All members of this family are selective for Na+ and blocked by amiloride. The mammalian homolog of degenerins (MDEG or MDEG1, now designated ASIC for Acid Sensing Ion Channels). Three are at least three distinct proteins in ASIC family: ASIC1 (identical with human BNAC2 or BNC2), expressed in brain and dorsal root ganglions (DRG) cells, is activated by pH <7.0. Applications: Suitable for use in ELISA. Western Blot, though not tested, may potentially be used as an application. Other applications not tested. Recommended Dilution: Western Blot: 1:1000-1:5000 using ECL., ELISA: 1:10,000-1:50,000. Coat ELISA plates at 1ug/ml with Control Peptide., Optimal dilutions to be determined by the researcher. Recommended Control Peptide: A0576-06: Acid Sensitive Ion Channel 3, Rat, CT, 21aa (ASIC3, DRASIC, Dorsal Root Ganglion ASIC) Control Peptide. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
