X-linked Ectodysplasin Receptor (XEDAR), also known as EDA2R and TNFRSF27, is an ~45kD transmembrane protein in the TNF receptor superfamily. Mature mouse XEDAR consists of a 138aa extracellular domain (ECD) with three tandem TNFR cysteinerich domains, a 21aa transmembrane segment, and a 138aa cytoplasmic domain. Within the ECD, mouse XEDAR shares 87% and 96% aa sequence identity with human and rat XEDAR, respectively. A 20kD fragment of the ECD can be shed by metalloprotease mediated cleavage. XEDAR binds selectively to the EDAA2 variant of Ectodysplasin (EDA), while the closely related receptor EDAR binds selectively to the EDAA1 variant. Other than a 2aa deletion in its TNF-like-domain, EDAA2 is identical to EDAA1. Mutations in both EDAR and EDA are associated with hypohidrotic ectodermal dysplasia (HED), a disorder of hair, tooth, and eccrine sweat gland morphogenesis. XEDAR itself is strongly associated with androgenetic alopecia (male hair loss). XEDAR is widely expressed, notably in embryonic basal epidermal cells and maturing hair follicles. Even though it does not contain a cytoplasmic death domain, XEDAR can associate with Fas and induce EDAA2 dependent apoptosis. Its transcription is directly induced by p53, and XEDAR mediated cell death is p53 dependent. XEDAR is downregulated in breast, colon, and lung cancers, particularly in cases with p53 mutations. XEDAR also plays a role in EDAA2 induced skeletal muscle degeneration and osteoblast differentiation. Source: Recombinant corresponding to aa1-138 mouse XEDAR expressed in NSO cell line. Molecular Weight: ~42.6kD, Endotoxin: ~1.0 EU/1ug (LAL) , Biological Activity:, Measured by its binding ability in a functional ELISA. When recombinant mouse XEDAR Fc Chimera is coated at 0.1ug/ml (100ul/well), the concentration of recombinant human EDAA2 that produces 50% of the optimal binding response is typically 3-15ng/ml. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile buffer. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
