The immunoregulatory cytokine tumor necrosis factor alpha (TNFa) recognizes two cell surface receptors. TNF-R1 (also known as CD120a) has an apparent molecular mass of 55/60kD and is constitutively expressed on all nucleated cells, whereas TNF R2 (also known as CD120b) with an apparent molecular mass of 75/80kD, is restricted to hematopoietic cells. TNF-R2 directs TNFa mediated activation and proliferation of T lymphocytes. An autocrine role for TNF and TNF-R2 is implicated in hematopoietic malignancies, and the TNF-R2 locus is a translocation target in several leukemias, suggesting a role for TNF-R2 in the development of certain leukemias. The p55 receptor extracellular domain was produced in insect cells (Sf9) infected with recombinant viruses. Source: Sf9 , Biological Activity: Binding of the ligand TNF was demonstrated by receptor binding competition of binding with radioactive TNF. Storage and Stability: Lyophilized powder may be stored at -20°C. Reconstitute to with sterile dH2O or buffer, aliquot and store at -20°C. Reconstituted product is stable for 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
