Restriction enzymes are used to recognize a specific sequence of nucleotides and cut double stranded DNA into smaller fragments at a specific location. Each restriction enzyme recognises different DNA sequences. NcoI is a Restriction Endonuclease purified from an Escherichia coli strain that carries the NcoI gene from Nocardia corallina. NcoI activity is not affected by Dam methylation, Dcm methylation and CpG methylation. The optimum reaction temperature is at 37 °C.
