Human Aurora kinase A (GenBank Accession No. NM_003600), (aa 2-403) with N-terminal HIS6 tag, MW 50kD, expressed in an Baculovirus infected Sf9 cell expression system.. Applications: Suitable for use in the study of enzyme kinetics, screening inhibitors, and selectivity profiling. Other applications not tested. Specific Activity: ~ 120U/ug. One unit is defined as the amount of enzyme that will phosphorylate 1pmol of Ser/Thr 1 peptide substrate per minute at pH 7.4 and 30°C , Assay buffer: 50mM HEPES, pH 7.4, 3mM MgCl2, 3mM MnCl2, 1mM DTT, 3uM Na orthovanadate, 0.5mM ATP, 4uM aurora B substrate, and 0.4ug/ml Aurora A kinase. Storage and Stability: Store at -70°C. Aliquots are stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
