Product information "Calyculin A, Highly Purified"
Calyculin A, a polyene phosphate, was initially isolated from the marine sponge Discodermia calyx. It is a very potent, cell-permeable PP-1 and PP-2 inhibitor (IC50: 2nM and 0.5-1nM respectively, and extremely specific for PP-1 and PP2A classes of protein serine/threonine phosphatase among other acidic, alkaline and protein phosphatases. Calyculin A is a polyene phosphate, a potent inhibitor (Ki ~ 0.1nM) with high specificity for the PP-1 and PP-2 classes of protein serine/threonine phosphatase. Solubility: Soluble in DMSO and EtOH, insoluble in water. Calyculin A must first be dissolved in an organic solvent such as DMSO, ethanol, acetone or methanol. The recommended solvent for stock solutions to be used for biological experiments is DMSO. Activity: The protein phosphatase inhibitory activity is tested on adipocyte cytosolic protein phosphatase activity, using myelin basic protein phosphorylated by protein kinase C as a substrate. An Okadaic acid-resistant phosphatase is inhibited with ID50 of 200nM, while 1uM exerts full inhibition of total phosphatase activity. Applications: Inhibition of protein serine/threonine phosphatases (PP-1 and PP-2) Potent non-phorbol type tumor promotion. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with DMSO and EtOH. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. Do not expose to sunlight or fluorescent lights for extended periods. Warning! This product is a suspect cancer hazard and tumor promoter. Gloves and mask should be worn when using this compound. Care must be taken to prevent contact through all routes of exposure.
