Product information "CD13/Aminopeptidase N, BioAssay(TM) ELISA Kit (Mouse)"
Alanine aminopeptidase, also known as ANPEP or CD13. is an enzyme that is used as a biomarker to detect damage to the kidneys, and that may be used to help diagnose certain kidney disorders. It is mapped to 15q26.1. Aminopeptidase N is located in the small-intestinal and renal microvillar membrane, and also in other plasma membranes. In the small intestine, Aminopeptidase N plays a role in the final digestion of peptides generated from hydrolysis of proteins by gastric and pancreatic proteases, and it is also thought to be involved in the metabolism of regulatory peptides by diverse cell types, including small intestinal and renal tubular epithelial cells, macrophages, granulocytes, and synaptic membranes from the CNS. Petrovic et al showed that CD13 was required for endothelial cell invasion in response to bradykinin. Inhibition of CD13 abrogated internalization of bradykinin receptor B2 and reduced endothelial cell motility. Sample Type: Cell culture supernates, serum and plasma (heparin, EDTA), Intended Use: For quantitative detection of Mouse CD13 in cell culture supernates, serum and plasma (heparin, EDTA). Sensitivity: <10pg/ml, Range: 312-20,000pg/ml, Specificity: Natural and recombinant mouse CD13, Test Principle: This mouse CD13 BioAssay(TM) ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from rat specific for CD13 has been precoated onto 96-well plates. Standards(NSO) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for CD13 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the mouse CD13 amount of sample captured in plate. Kit Components: 96-well plate precoated with mouse CD13 antibody, 1, Lyophilized recombinant mouse CD13 standard, 20ng/tube×2, Biotinylated mouse CD13 antibody, 130ul (dilution 1:100), Avidin-Biotin-Peroxidase Complex (ABC), 130ul (dilution 1:100), Sample diluent buffer, 30ml, Antibody diluent buffer, 12ml, ABC diluent buffer, 12ml, TMB color developing agent, 10ml, TMB stop solution, 10ml, Storage and Stability: Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
