Cayman's MPO Inhibitor Screening Assay Kit provides convenient fluorescence-based method for identifying novel inhibitors of the chlorination and peroxidation activities of myeloperoxidase (MPO). The chlorination assay uses the non-fluorescence APF. APF is selectively cleaved by hypochlorite (-OCl) to yield free fluorescein. which can be easily quantified using a fluorescence plate reader at excitation and emission wavelengths of 480-495 and 515-525 nm, respectively. The peroxidation assay is based on the two-electron oxidation of native MPO to MPO-I by hydrogen peroxide, followed by two successive one-electron reductions back to MPO by ADHP with an MPO-II intermediate. The reaction between hydrogen peroxide and ADHP yields resorufin, which can be easily quantified at excitation and emission wavelengths of 530-540 and 585-595 nm, respectively.Synonyms: MPO Inhibitor Screening Assay Kit.
