Product information "Matrix Metalloproteinase 3 (MMP-3) Activity Fluorometric Assay Kit"
Detection principle: This kit is detected by fluorescence resonance energy transfer (FRET) method. MCA and Dnp are connected to two ends on the natural substrate of MMP-3 enzyme. When MMP-3 protease does not cut the substrate, the two groups are close enough to undergo fluorescence resonance energy transfer, that is, Dnp can quench the fluorescence of MCA, resulting in undetectable fluorescence. When the substrate is cut by MMP-3 protease, the ends and ends of the polypeptide are separated, the two groups are separated, the fluorescence of MCA is no longer extinguished by Dnp, and the fluorescence of MCA can be detected, so that the enzyme activity of MMP-3 protease can be detected very sensitively through fluorescence detection. MCA has a maximum excitation wavelength of 325 nm and a maximum emission wavelength of 393 nm. Performance characteristics: Sample type: serum, plasma, animal tissue, cell Sensitivity: 0.03 U/L Detection range: 0.03-9.66 U/L Detection method: Fluorescence method Assay type: Enzyme Activity Assay time: 50 min Precision: Average inter-assay CV: 7.5-7.7%Average intra-assay CV: 2.0-4.0% Other reagents required: Normal saline (0.9% NaCl) Storage: -20? Valid period: 12 months. Protein function: Metalloproteinase with a rather broad substrate specificity that can degrade fibronectin, laminin, gelatins of type I, III, IV, and V, collagens III, IV, X, and IX, and cartilage proteoglycans. Activates different molecules including growth factors, plasminogen or other matrix metalloproteinases such as MMP9 (PubMed:11029580, PubMed:1371271). Once released into the extracellular matrix (ECM), the inactive pro-enzyme is activated by the plasmin cascade signaling pathway (PubMed:2383557). Acts also intracellularly (PubMed:22265821). For example, in dopaminergic neurons, gets activated by the serine protease HTRA2 upon stress and plays a pivotal role in DA neuronal degeneration by mediating microglial activation and alpha- synuclein/SNCA cleavage (PubMed:21330369). In addition, plays a role in immune response and possesses antiviral activity against various viruses such as vesicular stomatitis virus, influenza A virus (H1N1) and human herpes virus 1 (PubMed:35940311). Mechanistically, translocates from the cytoplasm into the cell nucleus upon virus infection to influence NF-kappa-B activities (PubMed:35940311). [The UniProt Consortium]
