Restriction enzymes are used to recognize a specific sequence of nucleotides and cut double stranded DNA into smaller fragments at a specific location. Each restriction enzyme recognises different DNA sequences. EcoRI is a Restriction Endonuclease purified from an Escherichia coli strain that carries the EcoRI gene from Escherichia coli RY 13. EcoRI activity is blocked by some combinations of overlapping CpG methylation. The optimum reaction temperature is at 37 °C.
