Growth/differentiation factors (GDF-1 to GDF-15) are members of the BMP family of TGF-beta superfamily proteins. They are produced as inactive preproproteins which are then cleaved and assembled into active secreted homodimers. GDF dimers are disulfide-linked with the exception of GDF-3 and -9. GDF proteins are important during embryonic development, particularly in the skeletal, nervous, and muscular systems. Intended Use: For the quantitative determination of Growth Differentiation Factor 8 (GDF-8) concentrations in cell culture supernates, tissue homogenates, serum, and plasma. Sensitivity: 5.32pg/ml, Range: ~31.3-2000pg/ml, Test Principle: This assay employs the quantitative sandwich enzyme immunoassay technique. A monoclonal antibody specific for mature GDF-8 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells, and any GDF-8 present is bound by the immobilized antibody. After washing away any unbound substances, an enzyme-linked monoclonal antibody specific for mature GDF-8 is added to the wells. Following a wash to remove any unbound antibody-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of GDF-8 bound in the initial step. The color development is stopped and the intensity of the color is measured. Kit Components: Microplate, 1x96wells, Conjugate (HRP), 1x21ml, Standard, 1x20ng, Assay Diluent, 1x11ml, Calibrator Diluent concentrate, 1x21ml , Wash Buffer concentrate, 1x21ml, Color Reagent A, 1x12ml, Color Reagent B, 1x12ml, Stop Solution, 1x6ml, Plate Sealers, 1x4 adhesive strips, Storage and Stability: Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
