A CYP26 inhibitor (IC50 = 0.004 µM), selective for CYP26 over CYP2A1, CYP2C11, CYP17, and CYP19 (IC50s = 9.1, 7.2, 2.6, and 1.3 µM, respectively), increases the levels of all-trans retinoic acid in plasma, skin, liver, fat, kidneys, and spleen in rats at 2.5 mg/kg, inhibits estrogen-induced vaginal keratinization in ovariectomized rats (ED50 = 1 mg/kg), an effect that can be reversed by AGN 193109, increases pinnal epidermal thickness and tail epidermal orthokeratosis percentage in mice from 0.6-2.6 mg/kg.
