A TXNIP inhibitor, inhibits TXNIP promoter activity in a reporter assay using INS-1 cells expressing human TXNIP (IC50 = 0.64 µM), decreases the levels of TXNIP in the same cells in a concentration-dependent manner, induces glucagon secretion from TC-1 mouse lung cancer cells and decreases glucagon-induced glucose production in primary mouse hepatocytes, decreases fasting blood levels of glucose, serum levels of glucagon and triglycerides, and hepatic levels of triglycerides in mice at 70 mg/kg per day, decreases blood levels of glucose and serum levels of glucagon in db/db mice but not in Txnip-/- mice.
