OICR-12694 is an inhibitor of Bcl-6. It binds to the BTB domain on Bcl-6 (Kd = 5 nM in a surface plasmon resonance (SPR) assay). OICR-12694 decreases transcription of Bcl-6-targeted genes in a reporter assay using SU-DHL-4 cells (EC50 = 89 nM). It reduces the proliferation of Karpas-422 large B cell lymphoma cells (IC50 = 92 nM).
