IRE1alpha kinase-IN-1 is an allosteric inhibitor of inositol-requiring enzyme 1alpha (IRE1alpha, IC50s = 160 and 80 nM for IRE1alpha kinase and ribonuclease activities, respectively). It is selective for IRE1alpha over a panel of 454 wild-type and mutant kinases at 1 µM. IRE1alpha kinase-IN-1 inhibits endoplasmic reticulum (ER) stress-induced IRE1alpha oligomerization in HEK293 cells (IC50 = 740 nM).
