Mouse TYR(Tyrosinase) ELISA Kit

This is a copper-containing oxidase that functions in the formation of pigments such as melanins and other polyphenolic compounds (PubMed:2517217). Catalyzes the initial and rate limiting step in the cascade of reactions leading to melanin production from tyrosine (PubMed:1537333, PubMed:2517217, PubMed:35469906). In addition to hydroxylating tyrosine to DOPA (3,4-dihydroxyphenylalanine), also catalyzes the oxidation of DOPA to DOPA-quinone, and possibly the oxidation of DHI (5,6-dihydroxyindole) to indole-5,6 quinone (PubMed:1537333) Assay Type: Sandwich. Test principle: The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Mouse TYR. Standards or samples are added to the appropriate microtiter plate wells then with a biotin-conjugated antibody specific to Mouse TYR. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Mouse TYR, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Mouse TYR in the samples is then determined by comparing the OD of the samples to the standard curve.
