The catenins are polypeptides that bind to the conserved cytoplasmic tail of cadherins and are required for cadherin function (2). a-catenin is a 102kD protein that is associated with the cadherin indirectly through its interaction with b-catenin and g-catenin (3). b-Catenin and its homolog plakoglobin associate directly with the cadherin (4). Thus, the cell-cell adherens junction is a structure composed of the transmembrane cadherin, which is associated directly with either b-catenin or plakoglobin, which in turn assoctiates directly with a-catenin. a- catenin then mediates the interaction between the cadherin-catenin complex and the actin cytoskeleton (5,6). a-catenin has been characterized as a tumor suppressor, suggesting that it may have a regulatory function in regulation to its structural role (7). b-Catenin links cadherin cell adhesion molecules to the cytoskeleton and has recently been implicated in the wnt/wingless signaling pathway and in the progression of neoplasia. b-Catenin associates with the tumor suppressor adenomatous polypopsis coli (APC) and functions as a co-activator of the TCF transcription factor family. It is regulated by glycogen synthase kinase (GSK) 3b, which phosphorylates and targets b-Catenin for ubiquitin-mediated proteolysis. Stabilization of b-catenin, either by wnt-mediated inhibition of GSK3b or by mutation (as found in numerous colorectal tumors and melanoma cell lines), results in activation of TCF responsive genes which are thought to play key roles in development and cancer progression. Applications: Suitable for use in Western Blot, Immunoprecipitation and Immunocytochemistry. Other applications have not been tested. Recommended Dilution: Western Blot: 0.1-2ug/ml. Detects b-catenin in an A431 total cell lysate. A431 total cell lysate was resolved by electrophoresis, transferred to nitrocellulose, and probed with anti-b-Catenin (0.1ug/ml). Proteins were visualized using a goat anti-rabbit IgG labeled with HRP and a chemiluminescence detection system., Immunoprecipitation: 4ug. Immunoprecipitates b-Catenin from 500ug of A431 membrane fraction lysate., Immunocytochemistry: 1-5ug/ml. Demonstrates positive immunostaining for b-catenin at cell-cell junctions of A431 cells fixed with 4% paraformaldehyde and permeabilized with 0.25% Triton X-100., Optimal dilutions to be determined by the researcher. Positive Control (Included): A0001-50: A431 Cells, Non-Stimulated in RIPA Buffer, Sample Buffer, Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, aliquot to avoid repeated freezing and thawing and freeze at -20°C. Aliquots are stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
