Virus-basierte Tools: Lentivirus, AAV und VSV
Virus-basierte Tools können zur Manipulation genetischen Materials und zur Untersuchung viraler Infektionen genutzt werden. Unser Partner BPS Bioscience bietet Infektionssysteme an, die auf Lentiviren, Adeno-assoziierten Viren (AAV) oder auf dem Vesicular stomatitis-Virus (VSV) basieren. Mit Lentiviren lassen sich z.B. stabil transfizierte Zelllinien erzeugen, sie besitzen eine hohe Infektionsrate (>90%), bieten hohe Level an anschließender Proteinexpression und können sowohl in vitro als auch in vivo angewendet werden. In unserem Blog erfahren Sie mehr über Lentiviren und mögliche Anwendungen. AAVs sind als virale Vektoren nützlich, weil sie ein „gene of interest“ von bis zu 5 kb Länge tragen können. Da sie zu einer stabilen Genexpression führen und sie aufgrund ihrer geringen Immunogenität sicher angewendet werden können, sind AAVs die Vektoren der Wahl für die Entwicklung vieler Gentherapien. Das umhüllte VSV stellt ein wertvolles Instrument zur Erforschung des Eintritts, der Replikation und der Zusammensetzung von Viren dar. VSV-G, das Glykoprotein des Virus, ist verantwortlich für das Anheften und Eindringen der VSVs in die Wirtszelle und wird häufig zur Pseudotypisierung von viralen Vektoren verwendet.
