Hydrogen Peroxide Detection Kit (HydroRed) is designed for the quantitative measurement of hydrogen peroxide (H2O2) in biological samples using N-Acetyl-3,7-dihydroxyphenoxazine, a known fluorogenic substrate for peroxidase and H2O2. The kit comes in a convenient 96-well or 5 x 96 well format, with enough HydroRed ADHP, H2O2 Standard, HydroRed HRP and assay buffer for 100 or 5 x 100 reactions. Figure 1: Mechanism of action of Hydrogen Peroxide Detection Kit (HydroRed). ADHP (N-Acetyl-3,7-dihydroxyphenoxazine) is a highly specific fluorogenic substrate of peroxidases. It does not naturally fluoresce until it is oxidized by a peroxidase in the presence of H2O2, turning into a red, fluorescent product named resorufin. In our assay, the HydroRed ADHP reacts with H2O2 in the presence of HydroRed HRP to generate the red-fluorescent oxidation product that provides a highly stable signal with an excitation/emission maximum at 530/585 nm. Hydrogen peroxide (H2O2) functions as a signaling molecule and a marker of oxidative stress. Under physiological conditions, H2O2 participates in redox signaling pathways that regulate cell growth, differentiation, apoptosis, and immune responses. However, excessive H2O2 production can overwhelm antioxidant defenses, leading to oxidative damage of lipids, proteins, and DNA. It is therefore implicated in cancer, neurodegenerative disorders, atherosclerosis, and aging. Furthermore, H2O2 levels in biological fluids can provide valuable diagnostic insights, such as indicating airway inflammation in asthma. Therefore, sensitive and reliable measurement of H2O2 is critical for understanding redox biology, enabling early disease detection, and guiding therapeutic development.
