Baculovirus qPCR Titration Kit

Baculovirus qPCR Titration Kit is designed for efficient extraction of baculovirus DNA and accurate quantification of baculovirus titers using qPCR (quantitative polymerase chain reaction). The kit is optimized to preserve double-stranded viral DNA and delivers high sensitivity while minimizing contamination and non-specific background signals. It includes viral DNA purification columns for rapid viral DNA , along with our proprietary Wavequant(TM) 2x qPCR Master Mix and gp64-specific primers for enhanced specificity to baculoviruses. Also included are positive and negative controls, as well as the 5-(and-6)-Carboxy-X-rhodamine (ROX) dye for generating a reliable standard curve. This kit includes 10 or 50 purification columns, for extracting baculovirus DNA, and enough reagents for 20 or 100 qPCR reactions (duplicates are recommended). Baculoviruses are large, double-stranded DNA viruses that primarily infect insects, most notably Autographa californica and Bombyx mori. These viruses have become widely used in biotechnology for the expression of recombinant proteins in insect cell systems due to their ability to produce high yields of recombinant proteins. A commonly used system is the baculovirus expression vector system (BEVS), which utilizes a modified baculovirus genome to introduce foreign genes into insect host cells such as Sf9, Sf21, or High Five cells, which were derived from the fall armyworm Spodoptera frugiperda. Determining the titer of baculoviruses during their production is crucial for optimizing protein expression, and this can be achieved using qPCR. The gp64 gene, which encodes the viral envelope glycoprotein GP64, is unique to baculoviruses and is present as a single copy in the viral genome. This makes it an excellent target for qPCR, providing an accurate marker to measure viral titer. The cycle threshold (Ct or Cq) value can be used to determine the viral copy number (VCN) by comparing it with a standard curve, and it can also help detect irregularities such as contamination or other copy number variations (CNVs).
