Inhibitor of DNA synthesis for alpha-type mating cells of S. cerevisiae. This synthetic peptide is the mating pheromone alpha factor. Alpha-factor pheromone arrests yeast in G1 of the cell cycle by inducing mating genes which alter the cell surface. Amino Acid Sequence (Linear): H-Trp-His-Trp-Leu-Gln-Leu-Lys-Pro-Gly-Gln-Pro-Met-Tyr-OH, Synonyms: alpha-Mating factor, Pheromone alpha factor, H-Trp-His-Trp-Leu-Gln-Leu-Lys-Pro-Gly-Gln-Pro-Met-Tyr-OH, CAS No: 59401-28-4, Molecular Formula: C82H114N20O17S1, Molecular Weight: 1684D, Appearance: White to off-white powder, Purity: >95% (HPLC, MS), Mass Identification: By MALDI-TOF DE mass spectral analysis, mass to be within 0.1% of exact molecular weight, Solubility: Freely soluble in water, methanol and acetonitrile, Recommended Concentration: ~1-75uM, Recommended Buffer: 0.1M sodium acetate, pH 5.2, Storage and Stability: Lyophilized powder may be stored at 4°C for short-term only. Stable for 12 months at -20°C. Reconstitute and store at -20°C. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer. To avoid condensation moisture, allow the vial to reach room temperature before opening. If the contents of a vial are not used immediately, blanket with nitrogen and store dry and dark at -20°C. Peptides in solution have a much more limited stability. They are also highly susceptible to proteolytic degradation. Microbial proteases may be excluded by the use of freshly distilled water and sterile filtration. Gloves are recommended to avoid chance contact with human proteases. Shipping Temperature: Most peptides are also stable lyophilized at ambient temperatures and do not require refrigeration for short periods during transportation, although extended exposure to warm temperatures should be avoided. Additional Reconstitution Notes: It is difficult to recover a peptide that has aggregated in the presence of salt. If sufficient peptide is available, test the solubility on a small sample first. Please note that DMSO can completely oxidize the free thiol groups of cysteine in short-chain peptides within minutes and should be avoided as a solvent for these peptides. Peptides containing Trp, Met, Cys, Asn or Gln require special care to avoid oxidation, use oxygen free water.
