T cell immunoglobulin and mucin domain-2 (TIM-2) is an ~50kD member of the TIM family of immune regulatory proteins. TIMs are type I transmembrane proteins with one Ig-like V domain and a Ser/Thr-rich mucin domain in their extracellular regions. Mature mouse TIM-2 consists of a 210aa extracellular domain (ECD), a 21aa transmembrane segment, and a 53aa cytoplasmic domain. Rat TIM-2, by contrast, possesses the same structural architecture as mouse but contains an additional 46aa within the mucin domain. Over comparable regions of the ECD, mouse and rat TIM-2 share 74% aa sequence identity. The isolated ECD forms dimers and tetramers in solution, and these structures suggest the possibility of TIM-2/TIM-2 cis interactions. TIM-2 is expressed on Th2 cells, B cells, oligodendrocytes, hepatocytes, hepatic stellate cells, and epithelial cells lining the bile duct and renal tubules. It is upregulated on Th2 cells during cellular activation and functions as a negative regulator of Th2 cell expansion and immune responses. TIM-2 binds to ligands expressed on activated B cells, macrophages, dendritic cells, and T cells. Its ligation by Semaphorin 4A induces tyrosine phosphorylation in the cytoplasmic domain of TIM-2. TIM-2 binding to ligand(s) on hepatocytes inhibits hepatocyte differentiation and proliferation. TIM-2 additionally plays a role in iron homeostasis by serving as an endocytic receptor for the heavy chain of Ferritin (H-Ferritin). Molecular Weight: ~23.5kD, Biological Activity: Measured by its binding ability in a functional ELISA. Immobilized Recombinant Mouse TIM-2 (0.5ug/ml 100ul/well) can bind H-ferritinwith an apparent KD~3 nM. Endotoxin: ~1EU/1ug (LAL) , Storage and Stability:, Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile buffer. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 3 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
