Full length recombinant corresponding to human PDE11A4 with an N-terminal GST tag, expressed in a Baculovirus infected Sf9 cell expression system. Genebank: BC112393. Specific Activity: As reported , Unit Definition: 1 unit is the amount of P4072-11D that will convert 1 pmole 3', 5'-cAMP to 5'-AMP per minute at 37°C. Assay Conditions: 10mM Tris-HCl, pH 7.4, 10mM MgCl2, 0.1mg/ml BSA, 0.05% Tween-20, 200uM cAMP and serial dilutions of PDE11A4 at 37ºC for 20 min. Quantified by 5 nucleotidase cleaving the 5-AMP product and releasing the phosphate group which is detected by Malachite Green Reagent. Applications: Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling. Recommended Dilution: Optimal dilutions to be determined by the researcher. Storage and Stability: Avoid repeated freezing and thawing. Store at -70°C. Stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
