Lymphocyte activation gene 3 (LAG-3) protein is a CD4-like protein and member of the immunoglobulin superfamily (IgSF) that binds with high affinity to MHC class II molecules. LAG3 is expressed in activated T cells and natural killer cells, and the protein functions as an inhibitory co-receptor that modulates T cell homeostasis, proliferation, expansion, activation, and function. LAG-3 is comprised of an N-terminal extracellular domain with four IgSF domains, a transmembrane domain, and a C-terminal cytoplasmic domain with a KIEELE motif required for modulation of T cell function. Elevated LAG3 expression in chronic lymphocytic leukemia (CLL) cells is associated with reduced treatment-free survival in patients with CLL. Co-administration of antibodies against both LAG-3 and PD-1 reduces the number and percentage of CLL cells in blood and spleen in a CLL-splenocyte adoptive transfer mouse model. SNPs in LAG3 are associated with susceptibility to multiple sclerosis. Cayman's LAG-3 Extracellular Domain (human, recombinant) protein consists of 433 amino acids, has a calculated molecular weight of 47 kDa, and a predicted N-terminus of Val29 after signal peptide cleavage. By SDS-PAGE, under reducing conditions, the apparent molecular mass of the protein is 50 to 60 kDa due to glycosylation.Synonyms: CD223, Lymphocyte Activation Gene 3 Protein.
