ATP citrate lyase is the primary enzyme responsible for the synthesis of cytosolic acetyl-CoA in many tissues. The enzyme is a tetramer (relative molecular weight approximately 440,000) of apparently identical subunits. It catalyzes the formation of acetyl-CoA and oxaloacetate from citrate and CoA with a concomitant hydrolysis of ATP to ADP and phosphate. The product, acetyl-CoA, serves several important biosynthetic pathways, including lipogenesis and cholesterogenesis. In nervous tissue, ATP citrate-lyase may be involved in the biosynthesis of acetylcholine. Source:, Recombinant corresponding to full length human ATP citrate lyase with N-terminal GST tag expressed in baculovirus expression system (NM_001096). Specific Activity: ~400pmole/min/ug. A, Assay Condition: 25ul of ACLY reaction is conducted in a buffer containing 40mM Tris, pH 8.0, 10mM magesium chloride, 2mM DTT, 2mM potassium citrate, 0.1mM CoA, 6-100ng human ACLY at 30ºC for 60 minute. Enzyme activity was assayed using ADP-Glo reagents. Applications: Suitable for use in enzyme kinetics, inhibitor screening and selectivity profiling. Other applications not tested. Recommended Dilutions: Optimal dilutions to be determined by the researcher. Storage and Stability: Aliquot to avoid repeated freezing and thawing. Store at -70°C. Aliquots are stable for 6 months at -70°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
