FO-35 is an ionizable cationic lipid (pKa = 7.67) that has been used in the generation of lipid nanoparticles (LNPs) for the delivery of mRNA in vivo. Intranasal administration of LNPs containing FO-35 and encapsulating reporter mRNA accumulate in nasal epithelial tissue in mice. Nebulized administration of FO-35-containing LNPs encapsulating Cre mRNA induces the expression of tdTomato in the lungs of ferrets engineered to express tdTomato upon Cre-mediated recombination..
