An ionizable cationic lipid, has been used in the generation of LNPs for the delivery of mRNA in vitro and in vivo, radiolabeled ALC-0307-containing, CD4 antibody-conjugated LNPs encapsulating mRNA encoding luciferase selectively bind to CD4+ T cells in vitro, intravenous administration of ALC-0307-containing, CD4 antibody-conjugated LNPs encapsulating Cre recombinase-encoding mRNA induce reporter gene expression in approximately 60 and 40% of CD4+ T cells in the spleen and lymph nodes, respectively, of mice.
