DBCO (dibenzocyclooctyne) PEG4 Amine is an excellent building block for developing DBCO-based labeling probes. The PEG4 arm significantly increases the compound's hydrophilicity. DBCO reagent is a common class of copper-free click chemistry labeling reagents. They exclusively react with azide-tagged biomolecules. The DBCO click chemistry is one of the most common strain-promoted alkyne azide cycloadditions (SPAAC). Early click chemistry reactions required copper as a catalyst. Copper is cytotoxic and thus limits the application of click reactions in cells and other biological samples. The SPAAC of DBCO eliminates the cytotoxicity problem caused by copper. The DBCO click reactions are bio-orthogonal, i.e. they can occur within organisms without interfering with native biochemical processes, and are now widely used in bioconjugation, labeling and chemical biology.
