CFP (mCerulean3) Lentivirus are replication incompetent, HIV based, VSV-G pseudotyped lentiviral particles that are ready to transduce almost all types of mammalian cells, including primary and non-dividing cells. These particles contain a CFP (cyan fluorescent protein) under the control of an EF1a promoter. The lentiviruses also transduce a puromycin selection marker (Figure 1). CFP expression and transduction efficiency can easily be verified and optimized via fluorescence microscopy or flow cytometry. CFP (mCerulean3) has an excitation wavelength of 433 nm, an emission wavelength of 475 nm, with an extinction coefficient of 34,000 M-1cm-1. Fluorescent proteins have been extensively used in cell biology studies. The most commonly used fluorescent proteins are derived from the Aequorea Victoria GFP (green fluorescent protein). Engineering of the chromophore-forming amino acids can result in different absorption and emission characteristics. CFPs (cyan fluorescent proteins) tend to be dimmer than GFP, but allow for blue-shifted fluorescence, giving scientists the flexibility to use multiple colors.
