DNA is isolated by proprietary modified guanidine thiocyanate techniques. The DNA is dissolved in 1X TE (10mM Tris, pH 8.0, 1mM EDTA) buffer. Quality Control: 1. The quality and purity of genomic DNA were tested by spectrophotometer and electrophoresis. A260/280 is between 1.8 and 2.0, A260/230 is > 2.0. (detected in 10mM Tris-Cl, pH 7.5), 2. Plant genomic DNA concentration is determined by pico green measurement while other genomic DNA concentration is determined by UV260 measurement., 3. RNase treatment to ensure the RNA contamination is eliminated., 4. Hind III digestion test: genomic DNA was successfully digested by Hind III. , 5. b-Actin expression was tested by PCR amplification for Genomic DNA or PCR ready genomic DNA., 6. All plant genomic DNA concentration was measured by pico green method to assure the true quantity.
