STEAP1 Knockout HCT116 Cell Line is an HCT116 human colorectal carcinoma cell line in which STEAP1 (Six Transmembrane Epithelial Antigen of Prostate 1, STEAP Family Member 1) is no longer expressed due to CRISPR/Cas9 genome editing. This cell line was generated by electroporation of HCT116 cells with ribonucleoprotein complexes of Cas9 and an sgRNA targeting STEAP1. The STEAP1 gene has a paralog, STEAP1B, encoded at a different locus of the same chromosome. Because of the high sequence homology between STEAP1 and STEAP1B, expression of both genes is lost in this cell line (both have an identical 1-base pair insertion at the site of sgRNA binding). This cell line has been validated by genomic sequencing. STEAP1 (six transmembrane epithelial antigen of the prostate 1), also called STEAP, belongs to the STEAP family of metalloproteinases, which are involved in iron and copper regulation and other processes. It is a cell surface protein expressed in several types of cancer, including prostate and colorectal cancers (CRC), where STEAP1 was identified as a cancer-specific cell-surface antigen. The upregulation of STEAP1 is linked to cancer cell proliferation, and higher STEAP1 expression levels are correlated with poor patient survival. The differential expression of STEAP1 between cancer cells and normal tissue makes STEAP1 a promising target for antibody-drug-conjugate (ADC), T cell engager (TCE), and CAR (chimeric antigen receptor)-T cell therapy development for the treatment of prostate and colorectal cancers.
