Product information "NKG2D/ NFAT Luciferase Reporter Jurkat Cell Line"
NKG2D / NFAT Luciferase Reporter Jurkat Cell Line is a Jurkat cell line generated by transducing the NFAT-Luciferase Reporter Jurkat Cell Line (BPS Bioscience #60621) with NKG2D Lentivirus (BPS Bioscience #83056) to overexpress human NKG2D (natural killer group 2D) (NM_007360.4). Expression of NKG2D was confirmed by flow cytometry. This cell line was functionally validated in a co-culture assay with MICA CHO Cell Line (BPS Bioscience #83074) by testing NFAT-dependent luciferase reporter activation, and its inhibition by the NKG2D blocker Tesnatilimab. Figure 1. Illustration of NFAT-dependent activation of the luciferase reporter in a co-culture assay using MICA CHO Cell Line and NKG2D / NFAT Luciferase Reporter Jurkat Cell Line. NKG2D (natural killer group 2D), encoded by Klrk1 (killer cell lectin-like receptor subfamily K, member 1), is an immune activating cell surface receptor involved in target cell recognition and killing by Natural Killer (NK) and CD8+ T cells. Its ligands (NKG2DLs) are commonly overexpressed in transformed and infected cells or in cells with damaged DNA, such as MICA and MICB (MHC class I-related chain A and B) or the ULBP family (UL-16 binding proteins 1-6). NKG2D plays an important role in the recognition and elimination of potentially dangerous cells by activating adaptive and innate immune cells. NKG2D is a homodimer, which binds to the DAP10 (DNAX-activation protein 10) homodimer, a specific adaptor molecule that possesses a YINM motif in the cytoplasmic region to transfer the activating signal. NKG2D and its ligands are promising immunotherapy targets. A deeper understanding of the NKG2D-NKG2DL pathway will open new therapeutic opportunities in cancer and autoimmune diseases.
