The IL-2 Responsive Luciferase Reporter Ba/F3 Cell Line is a murine Ba/F3 cell line engineered to express IL2RA (interleukin 2 receptor subunit alpha), also known as CD25, (hIL2RA, NM_000417.3). The cDNA was delivered by lentiviral transduction of human IL-15 Responsive Luciferase Reporter Ba/F3 cell line (#83548), which express both human CD122 (hIL2RB, NM_000878.5) and CD123 (hIL2RG, NM_000206.3) separated by a self-cleaving P2A peptide as well as a firefly luciferase reporter driven by STAT5 (Signal Transducer and Activator of Transcription 5) response elements located upstream of the minimal TATA promoter. After activation by IL-2, the endogenous transcription factor STAT5 binds to the response elements, inducing transcription of the luciferase reporter. This cell line has been validated to respond to human IL-15 and IL-2. Figure 1: Illustration of the mechanism of IL-2 Responsive Luciferase Reporter Ba/F3 Cell Line. IL-2 (interleukin 2) and IL-15 (interleukin 15) both bind to and signal through a complex composed of IL-2/IL-15 receptor beta chain (IL2RB, CD122) and the common gamma chain (IL2RG, CD132) of the receptor complex, while CD25 (IL2RA) specifically recruits IL-2 with high affinity. Ligand binding to the receptor activates downstream JAK (Janus Kinase) tyrosine kinases, which phosphorylate transcription factor STAT5. STAT5 phosphorylation triggers the formation of a homodimer and translocation to the nucleus, where it can activate the transcription of the Firefly luciferase reporter driven by STAT5 response elements present in the promoter. IL-2 is a key cytokine mainly produced by activated CD4+ and CD8+ T cells following antigen stimulation. Unlike IL-15, IL-2 is secreted as a soluble monomeric polypeptide that binds with high affinity to a trimeric receptor complex containing CD25 (IL2RA, interleukin 2 receptor subunit alpha), which is known to be transiently upregulated on activated T cells and constitutively expressed on regulatory T cells (Tregs). This interaction facilitates signaling through the shared CD122 (IL-2/IL-15 receptor, IL2RB) / CD132 (gamma-C, IL2RG) heterodimer present on target immune cells. This drives the expansion, survival, and differentiation of effector T cells and natural killer (NK) cells, while simultaneously maintaining Treg-mediated immune tolerance. IL-2 is a pioneering cytokine of interest in cancer immunotherapy and autoimmune disease treatment. While wild-type recombinant IL-2 (aldesleukin) has long been used to stimulate anti-tumor immunity, its narrow therapeutic window and propensity to preferentially expand immunosuppressive Tregs have led to the development of next-generation IL-2 analogs and "biased" variants designed to optimize therapeutic outcomes.
