AP1 Luciferase Reporter RAW264.7 Cell Line

The AP1 Luciferase Reporter RAW 264.7 Cell Line is a murine RAW 264.7 cell line that expresses the stably integrated firefly luciferase reporter under the transcriptional control of AP1 (activator protein-1)-responsive elements. This cell line has been validated to respond to mouse RANKL and PMA. It was also tested with CC-401 (an ATP-competitive small-molecule inhibitor of c-Jun N-terminal Kinase (JNK)) in the presence of mouse RANKL or PMA. The stress-activated protein kinase/c-Jun N-terminal kinase (SAPK/JNK) family of proteins includes mitogen-activated protein kinases (MAPKs) that are activated by stress, inflammatory cytokines, mitogens, oncogenes, and inducers of cell differentiation and morphogenesis. Upon activation of the SAPK/JNK pathway, MAP Kinase Kinases phosphorylate and activate JNKs. The activated JNKs translocate to the nucleus where they phosphorylate and activate transcription factors such as c-Jun, c-fos, or ATF (activating transcription factor). Upon phosphorylation, these transcription factors form homo- or hetero-dimers named AP1, which bind to activator protein-1 (AP1) response element to promote the expression of hundreds of proteins. AP1 plays a role in cell differentiation and proliferation and is dysregulated in several types of cancer, as well as in inflammatory and metabolic diseases. RAW 264.7 is a murine macrophage cell line that was transformed by the Abelson murine leukemia virus (A-MuLV) and is useful for studies involving immunoreactivity.
