5T4 CHO Cell Line

5T4 CHO Cell Line is a CHO-K1 cell line over-expressing the human 5T4 gene (accession number NM_006670.5), also known as TPBG (trophoblast glycoprotein), under the control of the cytomegalovirus (CMV) promoter. This cell line was generated by lentiviral transduction followed by puromycin selection and limiting dilution. Individual clones were screened based on 5T4 expression by flow cytometry to obtain this high-expressing cell line. 5T4, also known as TPBG or trophoblast glycoprotein, is an oncofetal antigen typically found on solid tumors and fetal trophoblasts. It is characterized by glycosylation at its N-terminus and a core containing several leucine-rich repeats. 5T4 is expressed in many different primary and metastatic cancers and has limited expression in normal adult cells. Due to its broad expression across a wide range of tumor types, including both solid tumors and hematological malignancies, 5T4 has emerged as a promising therapeutic target. Current therapies in development include 5T4-targeting CAR (chimeric antigen receptor)-T cells, vaccines, and antibody-drug conjugates (ADCs) to deliver drugs specifically to 5T4-expressing tumors.
